Mostly derived from polyclonal sources, secondary antibodies represent a group of antibodies which bind the whole Ig molecules or fragments of primary antibodies (monoclonal and polyclonal), such as the Fc or Fab regions. The presence of two Fab’ fragments is a result of a reduction reaction splitting the F(ab')2 fragment. The later is generated from a cleavage of the hinge region by the proteolytic enzyme pepsin, which releases a F(ab')2- and a pFc' fragment.
As the Fc domain is known to be the constant domain within the same animal class, only one type of secondary antibody is necessary to target several types of primary antibodies. Therefore there is no need to label numerous types of primary antibodies but only one type of secondary antibody. Otherwise, when selecting a secondary antibody, you need to choose the one which recognizes the host species as well as isotype of the primary antibody of interest i.e. choose an anti-rabbit secondary antibody to detect a rabbit monoclonal primary antibody.

Streptavidin is a purified tetrameric protein (53kDa) from Streptomyces avidinii. Each monomer (13kDa) can bind a molecule of biotin with a very high affinity. This binding property makes it usable in widespread applications.
The Assay procedure required first adding the biotin labelled secondary antibody followed by the streptavidin conjugated to a label such as HRP or fluorescent probes. This amplification step is particularly useful when you come across a low level of target of interest, which is non-detectable by secondary labelled antibodies alone.
