Muscle differentiation

Skeletal muscle regeneration relies on adult stem cells called satellite cells. These satellite cells are quiescent but can respond to injury or growth signals to regenerate tissue through re-entering the cell cycle. Activated satellite cells first proliferate and differentiate into myoblasts, which can then further differentiate into myocytes and fuse to form multi-nucleated myotubes. Myotubes then bundle together to form mature contractile myofibers.

Alternatively, pluripotent stem cells (ESCs and iPSCs) have been used as a source for skeletal muscle cells through directed differentiation providing a valuable tool for tissue engineering approaches.

Skeletal Muscle Differentiation Kit

Differentiate human pluripotent stem cells using a simple 3-step process of media change and cell passaging. The kit achieves typically 70% MF20-positive myotubes without the need for cell sorting or transfection. Options are also available for differentiating skeletal muscle progenitors to mature myotubes.

Differentiate your human iPS/ES cells into skeletal muscle cells in just 7 days producing highly pure populations of skeletal muscle cells using a temperature-sensitive Sendai virus.

The skeletal muscle cells produced display typical skeletal muscle morphology and markers, such as myosin heavy chain (MHC). Following day 7, the cells may be maintained in the maintenance medium best suited to your needs (see below for our range of muscle maintenance medium).
